Simple Biological Tests for Differentiation of Mycobacterium fortuitum-chelonei Complex

Abstract

Recent outbreaks of non-tuberculous mycobacterial infections caused by M. fortuitum-chelonei complex (referred to MFCC) suggested that species and subspecies differentiation of the organisms is epidemiologically important. In this study, a simple dichotomous key for presumptive identification of the species of the MFCC by determining reactions to four biological tests, arylsulfatase test, iron-uptake test, 5% NaCl tolerance test and crocin reaction, was described. Arylsulfatase and iron-uptake positive test provides a rapid distinction of M. fortuitum from M. chelonei. M. fortuitum was further divided into biotype 1(bioval fortuitum) and biotype 2(biovar peregrinum) by crocin reaction. M. chelonei was further divided into biotype 1(subsp. abscessus) and biotype 2(subsp. chelonei) either by 5% NaCl tolerance or crocin reaction. These results indicated that four biological tests, i.e., arylsulfatase test, iron-uptake test, 5% NaCl tolerance test and crocin reaction could provide reliable means of presumptive differentiating the MFCC.
